IGF-DES
Growth Factor — IGF-1 Analogue IGF-DES (Des(1-3)IGF-1) is a truncated variant of IGF-1 missing its first three N-terminal amino acids. This modification sharply reduces binding to IGF binding proteins, giving IGF-DES several-fold greater potency than native IGF-1, though with a shorter half-life than the longer-acting IGF-1 LR3 analogue. Structure: Des(1-3)-IGF-1 Potency: Markedly higher than native IGF-1 Half-Life: Short (minutes) — shorter than LR3
IGF-DES
Mechanism of Action
Like other IGF-1 analogues, IGF-DES binds the IGF-1 receptor (IGF1R), activating PI3K/Akt and MAPK/ERK signalling to drive cell proliferation, protein synthesis, and anti-apoptotic effects.
Reduced IGFBP Binding
Removing the first three N-terminal residues eliminates a key IGF binding protein (IGFBP) interaction site, leaving more free, bioactive peptide available to engage IGF1R compared to native IGF-1 — the basis for its markedly increased potency.
Short Duration of Action
Unlike IGF-1 LR3, which is engineered for an extended half-life, IGF-DES clears rapidly, producing a short, intense burst of IGF1R activation rather than sustained signalling — a distinction relevant to research protocol design.
