Mechanism of Action
MOTS-c translocates from mitochondria to the nucleus under metabolic stress, where it activates the AMPK pathway and regulates nuclear gene expression — a retrograde signalling mechanism unique to mitochondria-derived peptides.

Insulin Sensitivity
Promotes glucose uptake in skeletal muscle by activating AMPK and inhibiting the folate-methionine cycle, which impairs fatty acid oxidation. Mouse studies demonstrate reversal of diet-induced insulin resistance.

Exercise & Ageing
Physically active individuals show higher MOTS-c plasma levels. Exogenous MOTS-c improved exercise capacity in aged mice to levels comparable to young controls — suggesting a role as an exercise mimetic.
