Sermorelin
Mechanism of Action
Sermorelin binds the GHRH receptor on anterior pituitary somatotrophs, stimulating the same natural signalling cascade as endogenous GHRH to promote GH synthesis and pulsatile release.
Preserved Physiological Feedback
Because it stimulates rather than replaces the pituitary’s own GH output, Sermorelin preserves the somatostatin-mediated negative feedback loop, reducing the risk of pituitary desensitisation seen with supraphysiological GH dosing.
Historical Clinical Use
Marketed in the US as Geref for diagnostic testing of GH reserve and treatment of paediatric growth hormone deficiency, Sermorelin was discontinued commercially in 2008 for manufacturing reasons unrelated to safety or efficacy, but its clinical data remains a key reference point for GHRH-analogue research.
